So what if Rome was destroyed by the Samnites or some other peoples in the 300s BC, specifically before Alexander the Great and Phillip II and how would that effectively lead to the rise of Seleucus I Nicator as the King of Macedon and TTL's Alexander III Megas' analogue?

Well Seleucus was one of Alexander's generals but did he have any prominence before Alexander's conquests? If not it seems unlikely he could replace either Philip or Alexander as king as that would require overthrowing the dynasty.
If you want a greater Seleucidian dominance possibly avoid his assassination then according to his Wiki entry he would have had a decent chance of taking over Thrace and Macedonia.
Not sure why a destruction of Rome would affect that? However it would mean there was no Rome to defeat the empire later on. Whether and how long such a large empire could survive might be another question however. Given the potential conflict between Greek and Persian elements and the huge size of the empire especially since it lacks the logistical unification bonus the Roman one had thanks to the Med.
